ZHO-197 is a dual bacterial peptide deformylase and methionine aminopeptidase inhibitor with antibacterial activity. ZHO-197 exhibits an Escherichia coli peptide deformylase IC50 of 0.021 μM and a human peptide deformylase IC50 of 3.289 μM. ZHO-197 displays broad-spectrum antibacterial activity against gram-positive and gram-negative bacterial strains. ZHO-197 can be used for the research of bacterial infection[1].

In Vitro

ZHO-197 (0.0033-10 μM; 3 h) potently inhibits recombinant Escherichia coli peptide deformylase with an IC50 of 0.021 ± 0.003 μM and shows selective inhibition for bacterial over human PDF[1].
ZHO-197 (0-256 μg/mL; overnight) exhibits antibacterial activity against gram-positive and gram-negative bacterial strains, with MIC values of 32, 2, 128, 4, and 2 μg/mL against Escherichia coli BW25113, E. coli ΔtolC, E. coli DSM30083, Staphylococcus aureus DSM20231, and Bacillus subtilis 168, respectively, and an MIC90 of 1.87 μg/mL against Mycobacterium tuberculosis Erdman[1].
ZHO-197 (30-300 μM; 72 h) shows no cytotoxicity against human peripheral blood mononuclear cells[1].
ZHO-197 (64 μg/mL; overnight) acts as a dual inhibitor, effectively inhibiting peptide deformylase and methionine aminopeptidase in intact Bacillus subtilis[1].
