3-Hydroxypropionyl-CoA is a metabolic intermediate and enzyme substrate/product that functions in multiple metabolic pathways. 3-Hydroxypropionyl-CoA serves as a substrate for 3-Hydroxypropionyl-CoA dehydratase (HY-E71752), a product of 3-hydroxypropionyl-CoA synthetase, and a hydrolysis substrate for Ehd3p. 3-Hydroxypropionyl-CoA participates in autotrophic CO2 fixation, modified β-oxidation, and reverse hydration reactions. Accumulation of 3-Hydroxypropionyl-CoA inhibits fatty acid β-oxidation at the Fox2p site and induces metabolic toxicity. 3-Hydroxypropionyl-CoA can be used in studies of hematogenously disseminated candidiasis.

Biological Activity
3-Hydroxypropionyl-CoA acts as a substrate for recombinant 3-Hydroxypropionyl-CoA dehydratase (HY-E71752) from Metallosphaera sedula, which catalyzes its dehydration to acryloyl-CoA with an apparent Km of 60 μM[1].
3-Hydroxypropionyl-CoA causes Candida albicans SC5314 cells to show complete growth inhibition on butyrate/propionate medium when accumulated (due to Ehd3p deletion), demonstrating that this metabolite potently and specifically inhibits fatty acid β-oxidation[2].
3-Hydroxypropanoyl-CoA is identified as a minor synthetic side product of 4-nitrobutyryl-CoA production and a degradation product of the human GCD-catalyzed oxidation of 4-nitrobutyryl-CoA, detected via mass spectrometry with distinct m/z values and intensities depending on the electron acceptor used[3].
