3-Phenyl-2H-azirine-2-carboxylic acid serves as the E3 ubiquitin ligase ligand for ZSH-2117 (HY-174415), a covalent and selective EGFR PROTAC degrader with a DC50 of 45 nM. ZSH-2117 significantly inhibits cell proliferation and reduces the protein levels of AKT and ERK in the downstream EGFR signaling pathway. ZSH-2117 effectively suppresses tumor growth in Ba/F3-EGFRL858R/T790M/C797S xenograft mouse models[1].
