Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Preparation of Calmagite Staining Solution[3]
40 mg Calmagite dissolved in 100 ml water
2. Analysis of biological materials
Serum: 50 μL serum was added to each tube directly, mixed with 2 mL buffer and 0.5 mL EGTA solution, and made up to 4 mL with water. Finally 1 mL Calmagite solution was added for color development, In addition, two standards containing 1 and 1.5 μg Mg2+ were always run parallel to check the dependability of the earlier standard curve.
3. Evaluate and compare the magnesium levels in serum and saliva of individuals with tobacco habits with and without potentially malignant disorders[2]
3.1 Serum collection
Two millilitres of blood was collected in plain red vacutainers by venipuncture using aseptic measures. At 3500 rpm, the serum was separated by centrifugation for 10 min.
3.2 Unstimulated saliva collection
Three millilitres of unstimulated whole saliva sample was collected from the mouth directly into the Eppendorf on a single occasion to press their tongue against their palate so that freshly secreted saliva was collected in the sublingual region for 5 min. Saliva was centrifuged at 3500 rpm for 10 min to remove the excess mucus and unwanted particles.
3.3 Serum and saliva estimation
The estimation of magnesium in serum and saliva was done by calmagite method.
