Alkyne-PEG2000-GE11 is a functionalized PEGylated lipid conjugate composed of three modular components: alkyne (propargyl/alkyne linker) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and GE11 peptide (YHWYGYTPQNVI) (HY-P10128) as the terminal targeting/functional ligand. Alkyne-PEG2000-GE11 does not contain a lipid anchoring group but instead provides a terminal reactive site for post-assembly ligand or probe conjugation on PEGylated nanoparticles.

Biological Activity
Alkyne-PEG2000-GE11 is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against EGFR (HER1) on the surface of various epithelial tumor cells (lung cancer, breast cancer, colorectal cancer, head and neck cancer).
Composition and role of each component:
1. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. GE11 (GE11 peptide (YHWYGYTPQNVI)): Targeting ligand-a 12-amino acid peptide identified through phage display screening that selectively binds to epidermal growth factor receptor (EGFR/HER1) without activating its downstream signaling cascade. Enables receptor-mediated endocytosis into EGFR-overexpressing tumor cells while avoiding pro-proliferative signaling, making it a safer alternative to EGF for targeted delivery.
