Antitubercular agent-59
Antitubercular agent-59 is an orally active antitubercular agent with an IC50 of 51 nM against Mycobacterium tuberculosis H37Rv. Antitubercular agent-59 inhibits growth of Bedaquiline (HY-14881)-resistant Rv0678G12C mutant strains. Antitubercular agent-59 exerts bactericidal activity in chronic tuberculosis mouse models with 1-Aminobenzotriazole (HY-103389) coadministration.

Biological Activity
Antitubercular agent-59 (Compound 6k) potently inhibits the growth of Mycobacterium tuberculosis H37Rv whole cells with an IC50 of 213 nM[1].
Antitubercular agent-59 shows moderate metabolic clearance in rat hepatocytes and human liver microsomes[1].

Dosage:100 mg/kg
-
Administration:I.g.; daily 5 days per week; 4 weeks
-
Result:Reduced CFU counts to 5.18 log10 CFU/lung when treated with 1-Aminobenzotriazole (100 mg/kg).
Had a mean log10 CFU/lung of 5.97, which was not significantly different from that of untreated control when treated with 6k alone at 100 mg/kg.
