DOPE-PEG2000-CSTSMLKAC: surface functionalization of liposomes and lipid nanoparticles for tumor-targeted drug delivery and tumor imaging.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric barrier reducing opsonization and RES clearance, prolonging circulation time;
(3) CSTSMLKAC peptide; a cyclic tumor-homing peptide (cyclized via disulfide bond between terminal cysteines) that specifically binds to tumor vasculature or tumor cells, with the cyclic conformation enhancing structural stability, protease resistance, and binding affinity compared to linear peptides, directing nanocarriers to tumor sites for improved drug deposition.
