CellTracker Blue CMAC (solution) is a non-fluorescent cell membrane permeable dye. The chloromethyl groups of CellTracker Blue CMAC are enzymatically cleaved by intracellular glutathione (GSH) to generate a fluorescent product (blue fluorescence, Ex/Em: 360/460 nm). CellTracker Blue CMAC is suitable for long-term cell tracking (up to 72 hours) and cell proliferation studies, and can also quantify GSH levels.

Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs)>[2][3]
1. Dye preparation
Stock solution: protect from light and store in aliquots.
Working solution: Dilute the stock solution with serum-free medium. Preheat the staining working solution to 37 °C. The corresponding stock solution can be diluted according to the actual situation. Note that if the solvent is DMSO, the cytotoxicity of DMSO must be considered, and a solvent control should be prepared; if the solvent is pure water, the working solution needs to be filtered and sterilized before adding cells.
2. Cell labeling
Remove the cell culture medium and wash the cells with PBS.
Add the dye working solution and incubate at 37 °C for 30 minutes.
Remove the dye and wash 3 times with pre-warmed culture medium.
Change to fresh culture medium and continue culturing or experiment (fluorescence signal reaches peak value within 1-2 hours).
3. Detection
Fluorescence microscopy: Ex 360 nm, Em 460 nm.
