Tyrosinase (EC 1.14.18.1) (Polyphenol oxidase) is a rate-limiting enzyme that controls the production of melanin. Tyrosinase is mainly found in melanosomes synthesized by skin melanocytes. Tyrosinase can be used in research on Parkinson's disease, melanoma and pigmentation.

Product Information Optimal pH: 6.0-7.0 Theoretical molecular weight: 128 kDa (tetramer) Tyrosinase activity assay Enzyme solution preparation Dissolve in ddH2O at a concentration of 1 mg/mL. Further dilute to a concentration of 200-400 U/mL. Experimental method Adjust the spectrophotometer to 280 nm and 25°C. Add the sample to each colorimetric tube in the following order: 1mL 0.5 M Phosphate buffer, pH 6.5 1 mL 0.001 M L-Tyrosine 0.9 mL ddH2O Oxygen is introduced into the colorimetric tube through a capillary for 4-5 min to oxidize the reaction mixture. Place the colorimetric tube on the spectrophotometer and record the A280 at 4-5 min as the absorbance of the blank group. Add 0.1 mL of appropriately diluted enzyme and record the A280 at 10-12 min. Calculation 1U = the amount of enzyme that causes the absorbance at 280 nm to increase by 0.001 per minute.
