DB1255
DB1255 is an ERG inhibitor that binds to the minor groove of DNA, with KD values of 26 nM and 86 nM for the ATGA and AATT sequences, respectively. DB1255 targets and binds to specific DNA minor grooves (single GC/AT-rich sequences), forming a hydrogen bond network with G bases and AT pairs via its thiophene group and amidino group, respectively, thereby blocking the binding of transcription factors. DB1255 can be used in the research of leukemia and prostate cancer.

DB1255 (0.25-1.5 μM; 15 min) strongly and specifically binds to the minor groove of DNA and protects DNA fragments with a 265 bp 3'-end label from DNase I digestion[1].
DB1255 (at DNA molar ratios of 1:1 and 2:1) significantly increases the Tm and stabilizes the DNA double strand in a system containing 3 μM DNA hairpin oligonucleotide duplex[1].
At a molar ratio of DB1255 to DNA ranging from 0.25 to 2.0 (monitored in real time), DB1255 induces a strong positive circular dichroism signal and exhibits a 1:1 monomer binding mode in a DNA hairpin duplex (3 μM) system[1].
DB1255 (0.001-0.3 μM) exerts high-affinity monomer binding in a surface plasmon resonance biosensor system immobilized with 5'-biotinylated DNA, with KD values of 26 nM and 86 nM for sequences containing ATGA and AATT, respectively[1].
