Description

In Vitro

CTAP-1 (10 μM; 50 min incubation) labels a perinuclear, copper-dependent labile copper pool in NIH 3T3 fibroblast cells, with fluorescence intensity increasing ~2- to 3-fold in copper-supplemented medium, and staining colocalizing with mitochondria and the Golgi apparatus[1].
CTAP-1 (2 μM) fluorescence in copper-loaded NIH 3T3 fibroblast cells is reduced by ~50% upon treatment with the exogenous Cu+ chelator DTO, confirming the signal is copper-dependent[1].

Immunofluorescence[1]

Cell Line: mouse NIH 3T3 fibroblast cells
Concentration: 10 μM (standard staining; immunofluorescence colocalization); 150 μM CuCl2 (cell pre-culture)
Incubation Time: 50 min (37 °C, DMEM for standard staining; room temperature, PBS for immunofluorescence colocalization); 12 h (cell pre-culture with CuCl2)
Result: Produced a weak perinuclear staining pattern in basal medium.
Showed an ~2- to 3-fold stronger emission intensity in cells pre-cultured with 150 μM CuCl2 for 12 h.
Colocalized staining pattern with subcellular locations of the Golgi apparatus and mitochondria.
