Cyclo(Gly-{Tyr(PO3H2)}-Val-Pro-Met-Leu)
Cyclo (Gly-{Tyr (PO3H2)-Val-Pro-Met-Leu) is a binding peptide for the PI3K SH2 domain, as well as an inhibitory peptide for the mitogenic signal transduction-mediating domain. Cyclo (Gly-{Tyr (PO3H2)}-Val-Pro-Met-Leu) binds to the SH2 domain of the phosphorylated PI3K p85 subunit, and the spatial arrangement of its phosphotyrosine residue and methionine residue is associated with this interaction.

In Vitro
Cyclo(Gly-{Tyr(PO3H2)}-Val-Pro-Met-Leu) (compound 2) exists as a mixture of cis and trans isomers in methanol/water solution, with the trans isomer predominant in 75% methanol/water, and phosphorylation of the tyrosine residue does not significantly alter the peptide's overall solution conformation, with the trans isomer positioning the pTyr and Met side chains 16-18 Å apart and the cis isomer positioning them 7-14 Å apart[1].
Cyclo(Gly-{Tyr(PO3H2)}-Val-Pro-Met-Leu) binds strongly to the PI-3 kinase SH2 domain, but with lower potency than its linear phosphorylated counterpart[1].
