DHFR-IN-24
DHFR-IN-24, a benzothiazole derivative, is a dihydrofolate reductase (DHFR) inhibitor. DHFR-IN-24 has intrinsic antibacterial activity against both Gram-positive and Gram-negative strains. DHFR-IN-24 synergistically combines DHFR inhibition with photodynamic therapy (PDT) for enhanced antibacterial activity against multidrug-resistant pathogens.

Biological Activity
DHFR-IN-24 (compound 12) inhibits MRSA ATCC 33591, S. aureus ATCC 25923, S. epidermidis CICC 10436, A. baumannii Bio 34235, A. baumanniiBio 53272, E. coli ATCC 25922, and K. pneumoniae CICC 21611, with MIC values of 0.5 μg/mL, 0.5 μg/mL, 2 μg/mL, 16 μg/mL, 16 μg/mL, 4 μg/mL, and 8 μg/mL, respectively[1].
Photodynamic activation of DHFR-IN-24 enhanced its antimicrobial efficacy by up to 64-fold through efficient generation of reactive oxygen species (ROS) in MRSA[1].
DHFR-IN-24 possesses inherent antibacterial activity and exhibits significantly enhanced antibacterial activity upon photodynamic activation[1].
DHFR-IN-24 exhibits significant time- and concentration-dependent antibacterial effects against both Gram-positive and Gram-negative bacteria[1].
DHFR-IN-24 demonstrates potent light-enhanced antibiofilm activity against MRSA and A. baumannii[1].
DHFR-IN-24 does not show significant cytotoxicity against human embryonic kidney cells (HEK293T) and cervical cancer cells (HeLa)[1].
