3-Chymotrypsin-Like Protease

3-Chymotrypsin-like protease (3CLpro) is a critical enzyme in the SARS-CoV-2 life cycle, responsible for cleaving the viral polyprotein into 11 essential non-structural proteins required for replication. As a conserved cysteine protease with a highly similar structure across human coronaviruses, 3CLpro is a prime target for antiviral drug development. Inhibitors that bind tightly to its active site—either covalently or non-covalently—can block polyprotein processing and halt viral replication. Strategies to identify inhibitors include targeting the catalytic site with peptide-like and small molecule compounds, as well as exploring allosteric sites to develop non-competitive inhibitors.
