DMN-Tre is a conjugate of a solvatochromic fluorescent dye and trehalose. DMN-Tre takes advantage of the substrate promiscuity of the endogenous antigen 85 protein complex in mycobacteria to be metabolically integrated into the hydrophobic mycobacterial membrane. Once entering this hydrophobic environment, the linked DMN dye fluorescence is "turned on", enabling specific labeling. DMN-Tre can be used to reflect bacterial metabolic activity and support physiological studies of Mycobacterium tuberculosis.

Description

DMN-Tre is a conjugate of a solvatochromic fluorescent dye and trehalose. DMN-Tre takes advantage of the substrate promiscuity of the endogenous antigen 85 protein complex in mycobacteria to be metabolically integrated into the hydrophobic mycobacterial membrane. Once entering this hydrophobic environment, the linked DMN dye fluorescence is "turned on", enabling specific labeling. DMN-Tre can be used to reflect bacterial metabolic activity and support physiological studies of Mycobacterium tuberculosis[1][2].

In Vitro

Guide (The following are our recommended solutions. These are only Guide and should be modified according to your specific needs).
1.Preparation: Prepare or obtain DMN-Tre stock solution.
2. Incubation: Incubate samples with a final concentration of 100
M (for M. smegmatis) or up to 1 mM (for M. tuberculosis (Mtb)) of DMN-Tre.
3. Time: For M. smegmatis, 30 min is typical. For Mtb in sputum or cultures, labeling may take up to 16 hours or overnight.
4. Detection/Fixation: The cells can be imaged directly. For clinical sputum samples, this is followed by fixation (e.g., 4% formaldehyde or 2.5% glutaraldehyde) before analysis with a fluorescent microscope or flow cytometer.
