DOPE-PEG2000-CGKRK: surface functionalization of liposomes and lipid nanoparticles for tumor-targeted drug delivery and tumor imaging, with specific homing to tumor vasculature.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric barrier reducing opsonization and RES clearance, prolonging circulation time;
(3) CGKRK peptide; a tumor-homing pentapeptide identified by in vivo phage display that specifically binds to p32/gC1qR/HABP1, a mitochondrial protein aberrantly expressed on the surface of tumor cells and tumor vasculature endothelial cells, directing nanocarriers to tumor tissues for enhanced drug accumulation; the cysteine residue also provides a thiol group for conjugation.
