DOPE-PEG2000-CREKA
DOPE-PEG2000-CREKA (DOPE-PEG2000-Mal-Cys-CREKA) is an amphiphilic lipid-PEG-peptide conjugate consisting of DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the CREKA peptide (Cys-Arg-Glu-Lys-Ala), a tumor-homing peptide identified by in vivo phage display. DOPE-PEG2000-CREKA is used for tumor stroma-targeted drug delivery.

Biological Activity
DOPE-PEG2000-CREKA: surface functionalization of liposomes and nanoparticles for tumor stroma-targeted drug delivery and tumor imaging, homing to clotting products in tumor extracellular matrix.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric shield reducing opsonization and RES clearance, prolonging circulation time;
(3) CREKA peptide; a tumor-homing pentapeptide identified by in vivo phage display that specifically binds to fibrin-fibronectin complexes deposited in tumor stroma as a result of occult tumor-associated blood coagulation, directing nanocarriers to tumor extracellular matrix; this targeting is unique to tumors as fibrin-fibronectin complexes are not present in normal tissues.
