DOPE-PEG2000-K237 (DOPE-PEG2000-Mal-Cys-HTMYYHHYQHHL) is an amphiphilic lipid-PEG-peptide conjugate composed of DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the K237 peptide (sequence: HTMYYHHYQHHL). K237 peptide is a tumor-homing peptide identified by phage display that specifically binds to vasculature in tumor tissues. DOPE-PEG2000-K237 is used for tumor-targeted drug delivery via functionalization of liposomes and nanoparticles.

Biological Activity
DOPE-PEG2000-K237: surface modification of liposomes and lipid nanoparticles for tumor vasculature-targeted drug delivery and anti-angiogenic research.
Structural composition:
(1) DOPE; a phospholipid with two oleoyl chains that inserts into lipid bilayers as a hydrophobic anchor and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer chain that forms a steric barrier reducing opsonization and RES clearance, thereby extending blood circulation time;
(3) K237 peptide; a tumor-homing peptide identified through phage display screening that specifically recognizes and binds to angiogenic tumor vasculature, directing nanocarriers to tumor blood vessels for enhanced drug deposition at tumor sites and anti-angiogenic effects.
