DOPE-PEG2000-SP94
DOPE-PEG2000-SP94 (DOPE-PEG2000-Mal-Cys-SP94) is an amphiphilic lipid-PEG-peptide conjugate composed of DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the SP94 peptide (sequence: SFSIIHTPILPL), a hepatocellular carcinoma (HCC) -targeting peptide. DOPE-PEG2000-SP94 is used for HCC-targeted drug delivery through liposome and nanoparticle functionalization.

Biological Activity
Description
In Vitro
DOPE-PEG2000-SP94: surface decoration of liposomes and lipid nanoparticles for hepatocellular carcinoma (HCC) -targeted drug delivery, enabling selective accumulation in liver cancer tissues.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that forms a stealth layer reducing opsonization and prolonging circulation time;
(3) SP94 peptide; an HCC-specific targeting peptide identified by phage display that specifically binds to receptors on hepatocellular carcinoma cells (not normal hepatocytes), directing nanocarriers to liver cancer tissues for enhanced drug uptake by HCC cells and improved therapeutic efficacy with reduced toxicity to normal liver tissue.

Liver Cancer Modeling
Liver cancer can be classified into primary liver cancer and secondary liver cancer. Secondary liver cancer is the metastatic liver cancer. Primary liver cancer includes hepatocellular carcinoma (HCC), intrahepatic cholangiocarcinoma (ICC) and fibrolamellar HCC, of which HCC is the most common form, accounting for approximately 90% of primary liver cancers[1]. HCC mouse models include chemical agent-induced models, transplanted tumor models, and genetic engineered models.
