DOPE-PEG3400-TAT is a functionalized PEGylated lipid conjugate composed of three modular components: dioleoylphosphatidylethanolamine (DOPE) as the lipid anchoring group, PEG, and TAT peptide (YGRKKRRQRRR) (HY-P0281) as the terminal targeting/functional ligand. Biological Activity
DOPE-PEG3400-TAT is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against non-specific cell membrane penetration (heparan sulfate proteoglycans).
Composition and role of each component:
1. PEG: Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. TAT (TAT peptide (YGRKKRRQRRR): Targeting ligand-a classical cell-penetrating peptide (CPP) derived from the transactivation domain of HIV-1 Tat protein. Its highly cationic arginine-rich sequence enables efficient transmembrane transport through multiple uptake mechanisms (macropinocytosis, direct penetration). It lacks cell-type specificity but provides potent non-specific cellular uptake enhancement.
