NK2R

In Vitro

EB1002 (120 minutes) selectively activates human/mouse NK2R in COS-7 cells, without inducing signal transduction in human/mouse NK1R or NK3R receptors[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

In Vivo

EB1002 (40-325 nmol/kg s.c.; 2 nmol ICV; single dose, daily, every other day; 7/12/21 days) induces weight loss via increased energy expenditure and appetite suppression, improves insulin sensitivity, and has effects independent of leptin, Mc4r, and Ucp1[1].
EB1002 (s.c. single dose; s.c. daily, 7 days; i.c.v. single dose) reduces body weight via decreased food intake and increased energy expenditure, improves glucose metabolism, and preserves lean mass in obese Mus musculus[2]. Wait, no, the original text uses "obese mice" not the Latin name. Correcting to follow the rules strictly without altering content:\n\nEB1002 (s.c. single dose; s.c. daily, 7 days; i.c.v. single dose) reduces body weight via decreased food intake and increased energy expenditure, improves glucose metabolism, and preserves lean mass in obese mice[2].

Animal Model: C57Bl/6 NRj (male and female, ≥8 weeks old); B6.V-Lepob/JRj (ob/ob, male and female, ≥8 weeks old); B6.129S4-Mc4rtm1Lowl/J (Mc4r-knockout, ≥8 weeks old); B6N-Tacr2tm1Zpg (Nk2r-floxed, ≥8 weeks old); Ucp1-knockout (≥8 weeks old, thermoneutral housing); CD-1 mice (≥8 weeks old); diet-induced obesity model (60% HFD for ≥12 weeks)[1]
Dosage: 40 nmol/kg; 325 nmol/kg; 2 nmol (ICV)
Administration: s.c. (single dose); s.c. (daily, 7/12/21 days); s.c. (every other day, 21 days); ICV (single dose, 1.5 μl artificial CSF)
Result: Increased oxygen consumption, fatty acid oxidation, and core temperature; reduced food intake, respiratory exchange ratio (RER), and body weight in DIO mice (single 325 nmol/kg s.c.).
Reduced body weight (solely fat mass loss, lean mass spared), improved glucose tolerance, and increased insulin sensitivity (50% higher glucose infusion rate during clamp) in DIO mice (daily 325 nmol/kg s.c. for 7 days).
Reduced food intake, body weight, and blood glucose in ob/ob mice (325 nmol/kg s.c.).
Reduced food intake and body weight but not oxygen consumption or blood glucose in ob/ob mice (ICV 2 nmol).
Abolished effects in Nk2r-knockout mice. Reduced body weight, blood glucose, RER, and increased fatty acid oxidation, oxygen consumption, and body temperature in Ucp1-knockout mice at thermoneutrality (single 325 nmol/kg s.c.) compared to vehicle and pair-fed controls.

In Vivo:

Select the appropriate dissolution method based on your experimental animal and administration route.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 5 mg/mL (3.25 mM); Clear solution

This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
  • Protocol 2

Add each solvent one by one:  10% DMSO    90% (20% SBE-β-CD in Saline)

    Solubility: ≥ 5 mg/mL (3.25 mM); Clear solution

This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
