Eculizumab (Anti-Human C5, Humanized Antibody) is a long-acting humanized monoclonal antibody targeted against complement C5. Eculizumab inhibits the cleavage of C5 into C5a and C5b and inhibits deployment of the terminal complement system including the formation of membrane attack complex (MAC). Eculizumab prevents anti-ganglioside antibody-mediated neuropathy in mice. Eculizumab can be used in hemolysis studies.

Eculizumab (3-100 μg/mL) completely prevents MAC deposition, terminal axonal neurofilament loss, and damage to perisynaptic Schwann cells, and inhibits the increase in MEPP frequency, asynchronous twitching of muscle fibres, and the occurrence of 'silent' NMJs, and prevents CGM3/NHS-induced loss of contraction force in a concentration-dependent manner in the in vitro MFS model[1].

Flow cytometric analysis of 1X106 HepG2 cells with Eculizumab (HY-P9914, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG2 kappa Isotype Control (HY-P99002, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -
Immobilized Human Complement C5 Protein (Flag Tag) can bind Eculizumab. The EC50 for this effect is 3.17 ng/mL.

Effect of complement inhibition and FcγRII blockade on patient serum-induced platelet aggregation in HIMEA. Platelet aggregation induced by patients’ serum obtained during active disease (T2Pre) in the presence of (UFH, 100 IU/mL) was reduced by treatment with the anti-C5 antibody Eculizumab (0.35 mg/mL; 20 min), the C3 inhibitor compstatin (0.12 and 0.18 mg/mL), or the FcγRII-blocking antibody IV.3 (35 μg/mL). Results are expressed as the percentage of residual platelet reactivity relative to aggregation observed with UFH 100 IU/mL alone.
