Alentemol
Alentemol (U-68553B free base) is a potent dopamine receptor agonist that can cross the blood-brain barrier. Alentemol inhibits electrical impulse firing and dopamine release of dopamine neurons in the nigrostriatal pathway by activating presynaptic dopamine autoreceptors, without primarily relying on feedback regulation by postsynaptic receptors. Alentemol can be used for research on schizophrenia.

In Vivo
Alentemol (1.0 mg/kg; s.c.; single administration; 30 min) exhibits activity in entering brain tissue and reaching a certain intracerebral concentration in the normal male CF-1 mouse model[1].
Alentemol (0.1 μg/kg-0.1 mg/kg; s.c.; single administration; recording starts 10 min after administration and lasts for 20 min) significantly inhibits locomotor stimulation in the d-amphetamine-induced hyperlocomotion model in male CF-1 mice[1].
Alentemol (3 μg/kg-0.3 mg/kg; s.c.; single administration; 30 min) significantly reduces 3-MT levels in the striatum of male CF-1 mice in a normal model[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model:CF-1mice (male; 18-22 g) stimulated with 1 mg/kg d-amphetamine[1]
-
Dosage:0.0001, 0.001, 0.01, 0.1 mg/kg
-
Administration:s.c.; single administration; recording starts 10 min after administration and lasts for 20 min
-
Result:Potently and dose-dependently antagonized d-amphetamine-stimulated locomotor activity.
-
Animal Model:Upjohn CF-1 mice (male; 25-35 g)[1]
-
Dosage:0.003, 0.03, 0.3 mg/kg
-
Administration:s.c.; single administration; 30 min
-
Result:Significantly and dose-dependently reduced 3-methoxytyramine (3-MT) levels in the striatum, indicating inhibition of dopamine release.
