Description

In Vitro

The HW12 peptide can be successfully conjugated with MAL-PEG-PCD via a thiol-maleimide addition reaction to form HW12-PEG-PCD, and this result is confirmed by 1H NMR analysis[1].
The cellular uptake of FC-labeled HW12-linked mixed micelles (12 h) by MIA PaCa-2 cells is significantly lower than that of GE11-conjugated mixed micelles, indicating that HW12 cannot mediate EGFR-specific cellular uptake[1].
HW12-linked mixed micelles (60-500 nM GEM; 72 h) exhibit lower cytotoxicity against MIA PaCa-2 cells than GE11-linked mixed micelles, with an IC50 of 126.58 nM[1].

Cell Cytotoxicity Assay[1]

Cell Line: MIA PaCa-2 pancreatic cancer cells
Concentration: HW12-linked mixed micelles with GEM concentrations of 60, 500 nM
Incubation Time: 72 h
Result: Showed lower cytotoxicity than GE11-linked mixed micelles and unmodified micelles at all tested GEM concentrations.
Reached an IC50 value of 126.58 nM, which was 1.3-fold higher than the IC50 of GE11-linked mixed micelles (95.32 nM) and 1.1-fold higher than the IC50 of unmodified micelles (116.91 nM).
In Vivo

HW12-linked mixed micelles (40 mg/kg equivalent dose of gemcitabine; i.v.; once weekly; 3 weeks) deliver gemcitabine but show minimal orthotopic pancreatic tumor growth inhibition in athymic nude mice, with significantly lower efficacy than EGFR-targeted GE11-linked mixed micelles[1].
