Biological Activity
4-Hydroxyindole (0–200 μM, 24 h) inhibits amyloid β (Aβ1-42, 5 μM) fibrillization in a dose-dependent manner, with an IC50 of approximately 85 μM[2]. Kinetic studies shows that 4-Hydroxyindole (5–50 μM, 14 days) inhibits amyloid β aggregation during the elongation phase. Amyloid fibrillization consists of three phases: nucleation (lag phase), elongation phase, and equilibrium phase. At 5 μM, Aβ1-40 fibrillization was inhibited by approximately 60%, while at 25 μM and 50 μM, fibrillization was almost completely suppressed[2]. 4-Hydroxyindole (5–50 μM) dose-dependently blocks amyloid β-induced toxicity in PC12 cells[1].

Result:Dose-dependently protected PC12 cells from Aβ1-40 and Aβ1-42 toxicity. The minimum concentration of 4-Hydroxyindole required to significantly inhibit Aβ1-40 was 2.5 μM, while the toxic effects of both Aβ1-40 and Aβ1-42 were completely blocked at 50 μM 4-Hydroxyindole.

Result:In mice, the LD50 was 620 mg/kg (intracranial injection), showing lower toxicity than its oxidation product (LD50 was 320 mg/kg). Oxidation product of 4-Hydroxyindole induced behavioral abnormalities in mice and reduced blood glucose, cholesterol, and blood urea nitrogen (BUN) levels while increasing liver function enzymes (ALT, AST) and alkaline phosphatase (ALP) levels.

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (15.7 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 1.57 mg/mL (11.79 mM); Clear solution
This protocol yields a clear solution of ≥ 1.57 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (15.7 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
