DPyPE is a neutral phosphatidylethanolamine lipid composed of a polyisoprene alkyl chain with two pyridine-containing phosphine ligands. DPyPE is mainly used in liposome formulations and to enhance the efficiency of gene delivery in vitro and in vivo. For example, DPyPE can be mixed with cationic lipids such as VC1052 (HY-156616) (in a 1:1 ratio) to form the vaccine adjuvant Vaxfectin (HY-142998). DPyPE assists VC1052 in binding to negatively charged pDNA to form a uniform liposome complex by regulating the fluidity and stability of the liposome membrane.

Biological Activity
DPyPE, as a neutral auxiliary lipid, enhances the structural stability of liposomes, prevents excessive aggregation of cationic lipids, and may also assist the complex to enter antigen-presenting cells. DPyPE does not directly participate in transfection, but indirectly enhances the immune response by optimizing the properties of liposomes, making Vaxfectin an adjuvant focused on activating humoral immunity[1].

The preparation and experimental operation steps of liposome complexes in the literature are as follows[2]:
1. Liposome preparation
Cationic lipid VC1052 and neutral lipid DPyPE were dissolved in chloroform at a molar ratio of 1:1, mixed, and the solvent was evaporated by nitrogen flow in a sterile glass vial to form a dry lipid film.
Vacuum drying overnight to remove residual solvent, each vial contains 1.5 μmol of VC1052 and DPyPE.
Add 1 mL of sterile water for injection (SWFI) to each bottle and vortex at maximum speed for 5 minutes on a vortex mixer to form a liposome solution (concentration of 1.5 mM).
2. Preparation of pDNA-liposome complexes
Prepare liposomes (in SWFI) and pDNA (dissolved in 2× carrier) at 2 times the final concentration.
Slowly inject an equal volume of liposomes into the pDNA solution using a syringe at a pDNA (phosphate): cationic lipid molar ratio of 8:1, 4:1, or 2:1, while pushing at a constant speed using a 26-28 gauge needle.
Gently vortex for a few seconds (vortex mixer 4) to form a uniform liposome-pDNA complex (stable for several hours at room temperature and needs to be injected within 20 minutes to 1.5 hours).
3. Animal injection
Mouse: In the awake state, 50 μL of the complex was injected into the quadriceps femoris with a 28-gauge needle, containing 1-25 μg pDNA on each side of the muscle, and single or multiple booster injections were used.
Rabbit: After anesthesia, 300 μl of the complex was injected into the quadriceps femoris with a 22-gauge needle (or a needle-free injection device was used), with a dose of 150 μg pDNA.
4. Detection indicators
The anti-NP antibody titer in the serum was detected by ELISA, the antigen-specific plasma cells in the bone marrow were counted by ELISPOT, and the muscle transfection efficiency was evaluated by β-galactosidase activity assay.

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (3.11 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (3.11 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
