IRAK4-IN-5 is an interleukin-1 receptor-associated kinase 4 (IRAK-4) inhibitor with a Ki value of 2.8 nM. IRAK4-IN-5 interacts with non-conserved residues of IRAK-4, thereby conferring selectivity over related kinases. IRAK4-IN-5 can be used in the research of inflammatory lesions (atherosclerosis, arthritis).

IRAK4-IN-5 (compound 16) inhibits IL-1-induced NF-κB activation in primary human monocytes with an IC50 of 0.3 μM[1].
IRAK4-IN-5 potently binds purified IRAK-4 with a Ki of 2.8 nM, and exhibits 217-fold selectivity over IRAK-1 and 892-fold selectivity over TAK1[1].
