Biological Activity
Guide (The following is our recommended experimental protocol. This protocol is for reference only and should be modified according to your specific needs).
1. Preparation of CPM working solution
1.1 Preparation of stock solution
Freshly prepare CPM DMSO solution with a concentration of 4.0 mg/mL.
1.2 Preparation of CPM working solution
Dilute it 40 times in assay buffer to prepare working solution.
Note: Keep it on ice during the experiment.
2. Formal experiment
2.1 CPM reactivity experiment
a. Mix CPM working solution (30 μL) with 570 μL assay buffer.
b. Monitor the fluorescence signal as the temperature increases (excitation wavelength 387 nm, emission wavelength 463 nm).
c. Analyze the value of melting temperature (Tm) according to the Bolzmann equation.
2.2 Covalent interaction evaluation experiment
a. Mix 30 μL of CPM working solution with 570 μL of assay buffer (buffer pH 8.0) containing 50 mM glycine.
b. Incubate at 80 °C for 30 min in the dark.
c. Cool the sample to 20 °C and add lysozyme to a final concentration of 4 μM.
d. Monitor the change in fluorescence intensity after heating.
