CL 118326

CL 118326 is a potent, selective, competitive inhibitor of mammalian pancreatic phospholipase A₂ (PLA₂) and a weak antagonist of platelet-activating factor receptor (PAF receptor). CL 118326 competitively inhibits mammalian pancreatic PLA₂ (porcine: IC₅₀ = 1.55 μg/mL), and shows no activity against snake venom or bee venom PLA₂. CL 118326 inhibits PAF-induced and thrombin-induced platelet aggregation, as well as the release of leukotriene (LTC₄) and histamine from basophil-enriched leukocytes. CL 118326 can be used for research on inflammation and allergic reactions.

CL 118326 (5 min) inhibits thrombin-induced (IC₅₀ = 7.9 μM) and PAF-induced (IC₅₀ = 5.8 μM) rabbit platelet aggregation, but does not inhibit aggregation induced by arachidonic acid or ADP, and exhibits no PAF agonist activity^[1].
CL 118326 (70 min) inhibits histamine and LTC₄ release from human basophil-enriched leukocytes stimulated by antigens and ionophores, with IC₅₀ values ranging from 1.36 to 2.34 μg/mL^[1].

CL 118326 (10-20 μg; intradermal injection; single administration) dose-dependently inhibits skin capillary leakage in guinea pigs induced by porcine pancreatic phospholipase A₂, with the highest lesion area inhibition rate reaching 87.2% at the 10 μg dose^[1].
