AZD8701
Based on 1 Customer Validation
AZD8701 (IONIS-1063734) is an antisense oligonucleotide targeting FOXP3 in regulatory T cells (Tregs), with a human IC50 of 65.2 nM. AZD8701 binds to intronic sites of all FOXP3 pre-mRNA isoforms and mediates dose-dependent FOXP3 knockdown via free uptake. AZD8701 can be used in cancer-related research.

FOXP3 65.2 nM (IC50) |
In Vitro
AZD8701 (65 nM) potently knocks down FOXP3 in primary human Tregs (IC50 65 nM) via free uptake, reduces expression of key FOXP3 target genes by 25-50%, and impairs Treg suppressive function in vitro[1].
AZD8701 (0.001-10 μM; 9 days) potently reduces FOXP3 protein expression in primary human Tregs (EC50 = 44 nM, IC50 = 65.2 nM) and downregulates FOXP3-dependent immunosuppressive biomarkers CTLA4, ICOS, GITR, and CCR8 at 1 μM[2].
AZD8701 (1 μM; 9 days) strongly reduces FOXP3 protein expression in human thymic natural Tregs and effector Tregs subsets from healthy donor PBMCs[2].
AZD8701 reduces the suppressive function of human inducible Tregs in an in vitro effector T cell proliferation assay[2].

In Vivo
AZD8701 (6.25-50 mg/kg; s.c.; daily; 4 days) drives dose-dependent knockdown of FOXP3 mRNA and protein in human Tregs in humanized NSG mice[2].

Result:Promoted dose-dependent knockdown of FOXP3 mRNA in splenic human Tregs.
Promoted significant downregulation of FOXP3 protein levels in splenic, blood, and bone marrow human Tregs at 25 mg/kg, reducing the percentage of FOXP3+ CD4+ T cells in spleen.

Protocols
-
RNA extraction experimental
By lysing cells, releasing RNA, and removing impurities such as proteins and DNA, high-purity RNA products are finally obtained. The commonly used traditional method is the guanidine isothiocyanate/phenol/chloroform method (Trizol), which is suitable for a variety of animal materials including animal tissues, microorganisms, cultured cells, etc., and most plant materials.
-
Cell Cytotoxicity Assay
Cytotoxicity assays are usually based on the assessment of cell membrane damage, which can also be indirectly detected by measuring cell viability. Detection methods include MTT assay, CKK-8 assay, LDH assay and ATP assay, etc.
-
RNA interference technology
RNA interference (RNAi) is a cellular mechanism that inhibits gene expression by suppressing gene transcription or activating RNA degradation. This mechanism was discovered in plants in 1998 by Andrew Fire and Craig Mello. Today, this phenomenon can be observed in almost all eukaryotes, including protozoa, flies, nematodes, insects, parasites, and mammals.
