Dexamethasone acetate (Dexamethasone 21-acetate) is the acetate form of the glucocorticoid Dexamethasone (HY-14648). Dexamethasone acetate has a longer duration of action than Dexamethasone. Dexamethasone acetate can accumulate at inflammatory sites, in mouse liver and mouse lung via nanostructured lipid carriers. Dexamethasone acetate can be used to prepare topical formulations for studies related to sensorineural hearing loss, γ-carrageenan-induced paw edema, chronic active hepatitis, pulmonary diseases, and inflammation after vitreoretinal surgery.

In Vitro
Dexamethasone acetate (28.2 mg particles in 150 mL water; 37 days; 5 separate samples in 45 mL water; 26 days) encapsulated in PLGA-magnetite nanoparticles releases a total of 37.8% of particle weight as Dexamethasone equivalent over 37 days in aqueous solution, with 2% to 23% of particle weight released over 26 days in replicate experiments[1].
Dexamethasone acetate (2 h; 48 h) loaded solid lipid nanoparticles exhibit an initial burst release of ~68% over 2 h followed by gradual release over 48 h in pH 7.4 phosphate buffer saline at 37°C[4].

In Vivo
DA-NLCs (3 mg/kg; i.v.; single dose) deliver Dexamethasone acetate (DA) to inflamed pleural sites at concentrations 8.3 times higher than free DA, with a 10.6-fold larger AUC0-t and prolonged residence time at the inflammatory site[2].
DA-NLCs (7 mg/kg; i.v.; single dose) preferentially accumulate in mouse liver and lung tissues, achieving 1.8- to 2.6-fold larger AUC0-ts than free DA, and prolong DA residence time in plasma[2].
Dexamethasone acetate-loaded poly(ɛ-caprolactone) nanofibers (200 µg; intravitreous implantation via transscleral injection; single administration) exhibit excellent ocular biocompatibility and safety in the Rattus norvegicus eye, preserving retinal and choroidal structure and photoreceptor function over 10 days following intravitreous implantation[3].
Dexamethasone acetate (10 mg/kg; i.v.; single dose) delivered via solid lipid nanoparticles exhibits remarkable lung-targeting in healthy Kunming mice, with a 17.8-fold higher lung AUC0-t and 7.10 relative overall targeting efficiency compared to the free drug solution, while reducing uptake by liver and spleen macrophages[4].
