CIM-834 is an orally effective inhibitor of SARS-CoV-2 membrane protein. CIM-834 can prevent the assembly of infectious virus particles without inhibiting the synthesis of viral RNA. CIM-834 can reduce the viral titer in the lungs of SCID mice infected nasally with SARS-CoV-2, block the spread of SARS-CoV-2 among Syrian hamsters, and inhibit the replication of SARS-CoV-2 (including variants) and SARS-CoV. CIM-834 can be used in related research on COVID-19.

In Vivo

CIM-834 (100 mg/kg; p.o.; once or twice daily for 3 consecutive days) reduces the titer of infectious SARS-CoV-2 in the lungs of SCID mice^[1].
CIM-834 (100 mg/kg, p.o., twice daily for 4 days, in combination with ritonavir 50 mg/kg, p.o., twice daily) reduces lung SARS-CoV-2 titers in Syrian hamsters^[1].

Animal Model:

CB-17/Icr-Prkdcscid/scid/Rj SCID mice (male, 7-9 weeks old, intranasal infection with SARS-CoV-2 B.1.351)^[1]
Dosage:

100 mg/kg (once daily; twice daily)

p.o.; once daily; 3 days; p.o.; twice daily; 3 days
Result:

Significantly and dose-dependently reduced the infectious virus titer and viral RNA load in lung tissue.
Reduced the infectious virus titer in the mouse lungs by more than 3 log₁₀ within 48 hours administration after infection.

Syrian hamsters (female, 8-10 weeks old, intranasal infection with SARS-CoV-2 USA-WA1/2020)^[1]
Dosage:

100 mg/kg (CIM-834); 50 mg/kg (ritonavir)
Administration:

p.o.; twice daily; 4 days (CIM-834 co-administered with ritonavir)
Result:

Reduced lung viral RNA levels by 3.4 log₁₀ and infectious viral titres by greater than 4 log₁₀.
Resulted in nearly normal lung parenchyma with only very focal perivascular inflammation.
Prevented transmission of SARS-CoV-2 from treated index hamsters to untreated sentinel hamsters.

Stock Solution Working Solution

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 2.5 mg/mL (5.05 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Protocol 2

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

Solubility: ≥ 2.5 mg/mL (5.05 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

Protocol 3

Add each solvent one by one: 10% DMSO 90% Corn Oil

Solubility: ≥ 2.5 mg/mL (5.05 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
