ML089 is an orally active inhibitor of phosphomannose isomerase (PMI) and PHOSPHO1, with an IC50 value of 1.3 μM against human PMI and an IC50 value of 0.8 μM against PHOSPHO1. ML089 blocks the catabolism of mannose-6-phosphate, directs mannose-6-phosphate to participate in protein glycosylation, increases mannose incorporation into cellular proteins, and reduces tritiated water production from mannose-6-phosphate. ML089 inhibits PMI in living cells. ML089 can be used in research related to congenital disorder of glycosylation type Ia (CDG-Ia).

ML089 (serially diluted concentrations; 20 min) potently inhibits recombinant human phosphomannose isomerase with an IC50 of 1300 nM[1].
ML089 inhibits recombinant human phosphomannomutase 2 with an IC50 of 83 μM, showing 69-fold selectivity for its primary target phosphomannose isomerase[1].
ML089 inhibits PHOSPHO1 with an IC50 of 800 nM[1].
ML089 (12.5-50.0 μM) increases 3H-mannose incorporation into proteins in human hepatoma C3A cells in a dose-dependent manner, with fold increases of 1.9 at 12.5 μM, 2.3 at 25.0 μM, and 3.2 at 50.0 μM, without detectable cellular toxicity[1].

This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
