L-Ethionine (L-ETH) is an amino acid derivative. L-Ethionine serves as a substrate for ATP:L-methionine S-adenosyltransferase, which targets its L-stereoisomer. L-Ethionine blocks the synthesis of N6-dimethyladenosine in rat liver tRNA/4S RNA via its major hepatic metabolite S-adenosylethionine, inducing methyl-deficient rat liver tRNA/4S RNA. L-Ethionine can be used in studies of liver cancer and hepatocarcinogenesis mechanisms.

Biological Activity
L-Ethionine (L-ETH) (10 mM; 24-48 h) elevates AFP mRNA levels up to eightfold and albumin mRNA levels up to threefold in primary Balb/cJ mouse hepatocyte cultures after 48 hours of treatment with 10 mM L-ethionine[3].

L-Ethionine (10 mg; i.p.; single injection) induces a 10- to 30-fold greater increase in hepatic α-fetoprotein mRNA levels in Balb/cJ mice compared to Balb/cCRBL, C3H/He, and C57BL/6 mice, while leaving hepatic albumin mRNA levels unaffected[3].
L-Ethionine (10-2500 mg/kg; i.p.; single dose) exhibits very low acute toxicity in male Swiss mice, with no mortality observed at doses up to 2500 mg/kg and only 1 death at 640 mg/kg over a 7-day observation period[1].
