Description

LY127210 free base is an orally active peripheral arterial vasodilating antihypertensive agent. LY127210 free base interferes with the baroreflex mechanism, exerts direct myocardial bradycardic activity, has no α or β receptor blocking properties, and acts via all conventional administration routes. LY127210 free base reduces vascular resistance, cardiac output, heart rate, left ventricular end-diastolic pressure, regional vascular resistance, as well as blood flow in the skin and cerebral cortex. LY127210 free base can be used in research related to hypertension[1][2].

In Vivo

LY127210 (10 mg/kg; i.p.; single dose) free base reduces mean arterial pressure by 35% in chloralose-anesthetized spontaneously hypertensive rats via a 26% reduction in systemic vascular resistance and a smaller reduction in cardiac output, without inducing reflex tachycardia[1].

Animal Model: SHR (male, 16-18 weeks old, 280-330g, anesthetized with methohexital followed by α-chloralose infusion)[1]
Dosage: 10 mg/kg

Produced non-significant decreases in cardiac output, stroke volume, and LVdP/dt max at 5 minutes post-dosing.
Maintained all hemodynamic changes for at least 20 minutes post-dosing.
