Milbemycin A3 is a 16-membered macrocyclic lactone compound found in the soil bacterium Saccharopolyspora hygroscopicus subsp. aureolacrimosus. Milbemycin A3 enhances the opening of glutamate- and GABA-gated chloride channels and exhibits insecticidal activity. Milbemycin A3 can be used in insect resistance-related research.

Biological Activity
Milbemycin A3 shows potent acaricidal and nematocidal activities as the major active component of the milbemycin A3/A4 mixture[1].
Milbemycin A3 (8 days) is eliminated of C5-O-methylated milbemycin byproducts and shows an increased yield of milbemycin A3/A4 to 2237 μg/mL in Streptomyces bingchenggensis BCJ13 with deleted milD gene; meanwhile, in strain BCJ36 with combined deletion of milD and nanLD genes, Milbemycin A3 is eliminated of both C5-O-methylated milbemycin byproducts and nanchangmycin[3].
Milbemycin A3, when analyzed in Streptomyces bingchenggensis BCJ36 (five successive subcultivations; 8 days per fermentation), shows genetic stability, maintaining elimination of unwanted byproducts[3].
