Description

miR-21-IN-2 is a high-affinity miR-21 inhibitor. miR-21-IN-2 binds to the apical loop region of the pre-miR-21 hairpin, inhibits Dicer-mediated processing, and thereby blocks the biosynthesis of mature miR-21. miR-21-IN-2 can be used for the research of microRNA-related diseases[1].

In Vitro

miR-21-IN-2 (Compound 1) (0.05-500 μM; 30 min) specifically binds to the 2-AP-labeled truncated 29-mer miR-21-hp RNA, with a Kd value of 2.3 μM as determined by 2-aminopurine fluorescence titration[1].
miR-21-IN-2 (0-250 μM; 30 min) binds to the 5′-Cy5-labeled truncated 29-mer miR-21-hp RNA, and the Kd value is determined as 3.2 μM by fluorescence intensity assay[1].
miR-21-IN-2 (0.1-100 μM; 0-2 h) inhibits Dicer-mediated cleavage of full-length pre-miR-21 RNA, with an inhibition rate of 47% at 1 μM, and exhibits dose-dependent activity in the range of 0.1 to 100 μM[1].

To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    90% Corn Oil

    Solubility: 2.5 mg/mL (5.12 mM); Suspended solution; Need ultrasonic

    This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
