N-Acetyl-L-proline undergoes stereoselective hydrolysis by purified N-acyl-L-proline-acylase derived from Comamonas testosteroni DSM 5416 in a system at 30 ℃ and pH 7.0. This enzyme has a specific activity of 85.2 U/mg, a KM value of 5 mM for the substrate, and enables complete conversion of the substrate after continuous incubation for 2 h[2].
N-Acetyl-L-proline derivatives with modified carboxyl groups show no catalytic reactivity toward purified N-acyl-L-proline-acylase from Comamonas testosteroni DSM 5416, which verifies that free carboxyl group is essential for substrate-enzyme binding[2].
