Description

NCR247 is a defensin-like nodule cysteine-rich (NCR) peptide and haem sequesterer. In Vitro

NCR247 (~116-120 s) binds haem with nanomolar affinity, with cysteine residues, the C10P11 motif, and tyrosine residues contributing to high-affinity binding; the D-enantiomer and M. sativa ortholog retain equivalent nanomolar haem-binding affinity[1].
NCR247 (5 μM; 16 min) sequesters haem and inhibits its intrinsic peroxidase activity, rendering the haem biologically inaccessible[1].
NCR247 (2-4 μM; 30-60 min) induces an iron starvation response in S. meliloti by increasing expression of iron uptake genes and reducing expression of rirA; this response requires uptake of NCR247 into the bacterial cytoplasm via BacA and is mediated by the Irr regulatory pathway[1].
NCR247 (2 μM; 30-60 min) drives increased iron import into S. meliloti, resulting in elevated total cellular iron content; this response is dependent on the Irr regulatory pathway[1].
L-NCR247 and D-NCR247 (2.5-20 μM; 24-48 h) inhibit growth of haem-auxotrophic bacterial pathogens H. influenzae and P. gingivalis, with D-NCR247 more potent than L-NCR247; haem addition reverses this growth inhibition, confirming haem sequestration as the mechanism of action[1].
L-NCR247 and D-NCR247 (25-150 μM; 6 h) inhibit Toxoplasma gondii plaque formation on human foreskin fibroblasts in a concentration-dependent manner, with haem binding required for the activity[1].

NCR247 (endogenous expression; 2 μM; in vitro treatment) is essential for effective legume-rhizobium symbiosis, as its depletion reduces nodule iron content and disrupts normal nodule development, while exogenous NCR247 boosts iron uptake in symbiotic bacteroids[1].
