OX2-2303 is a potent, selective orexin-2 receptor (OX2R) antagonist. OX2-2303 exhibits excellent binding affinity towards OX2R (Ki = 0.1 nM), and shows >890-fold selectivity over OX1R. OX2-2303 can be used as a positron emission tomography (PET) radioligand for central nervous system (CNS) research.

OX2-2303 (compound 1) exhibits no significant off-target activity across a panel of 59 major CNS targets, with exception of low affinity binding to sigma-2 (Ki = 2681 nM) and NR2B receptors (Ki = 9475 nM)[1].
OX2-2303 shows favorable tracer integrity, with no appreciable radiometabolites detected in saline, serum (mouse, rat, nonhuman primate, and human), and liver microsomes[1].
