PF-03475952 induces cancer cell apoptosis, inhibits LPS (HY-D1056)-induced leukocyte cytokine release and cancer metastasis, and reduces CD44 expression on circulating CD3+ lymphocytes in cynomolgus monkeys. In Vitro

PF-03475952 (0.0095-10000 ng/mL) binds potently and selectively to recombinant human and cynomolgus monkey CD44-Ig fusion proteins with EC50 values of 0.017 µg/mL and 0.015 µg/mL, respectively[1].
PF-03475952 (60 min) potently inhibits the interaction between recombinant CD44-Ig and HA with an IC50 of 0.41 µg/mL[1].
PF-03475952 (24 h) induces dose-dependent loss of cell surface CD44 from human CD14+ monocytes (IC50 = 3.3 µg/mL), human CD19+ B cells (IC50 = 2.4 µg/mL), human CD3+ T cells (IC50 = 2.6 µg/mL), and cynomolgus monkey CD3+ T cells (IC50 = 1.1 µg/mL) in whole blood[1].

Reduced CD44 expression on CD3+ lymphocytes below 50% of baseline for >504 hours at 100 mg/kg.
Achieved approximately 50% reduction in cell surface CD44 at serum levels of approximately 0.17 µg/mL for 1 mg/kg and 10 mg/kg dose groups.
Restored CD44 levels to baseline by day 22 (504 hours) in 1 mg/kg and 10 mg/kg dose groups.
