Phe-Lys(Trt)-PAB acts as a lysosomally cleavable substrate for cysteine protease cathepsin B, with cleavage enabling linked anticancer drug release via self-immolative spacer . Phe-Lys(Trt)-PAB can be used for synthesis of ADCs.

Phe-Lys(Trt)-PAB linker within human serum albumin-conjugated prodrug HSA-16 is efficiently cleaved by cathepsin B at pH 5.0 and 37 °C, releasing free Doxorubicin (HY-15142) and Paclitaxel (HY-B0015) with near-complete cleavage observed after 20 h[1].
