Pinostrobin chalcone (48 h) exhibits cytotoxic activity against MDA-MB-231 and HT-29 cancer cell lines, with IC₅₀ values of 55.41 µM and 83.54 µM, respectively^[1].
Pinostrobin chalcone (48-72 h) exhibits significant cytotoxic activity against KB, MCF7, and CaSki human cancer cell lines with IC₅₀ values of 6.2, 7.3, and 7.7 µg/mL, respectively, and also shows cytotoxicity against normal MRC5 cells with an IC₅₀ of 7.9 µg/mL^[2].
Pinostrobin chalcone docking predicts binding to human Janus kinase (JAK2) via hydrogen bonds mediated by its methoxy and hydroxyl groups, with the 3-position OH group forming a hydrogen bond with Asn859^[1].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Live/dead viability and cytotoxicity staining assays are based on the simultaneous detection of intracellular esterase activity in metabolically active (viable) cells and membrane integrity loss in non-viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
