Integrin alpha V beta 8

Integrin alpha V beta 8 pairs CD51 with the beta-8 subunit and, like alpha V beta 6, activates latent TGF-beta-1 through its RGD motif. Distinctively, the beta-8 cytoplasmic tail couples poorly to the cytoskeleton, so TGF-beta release occurs in a protease-dependent manner rather than by mechanical force. The receptor is expressed on dendritic cells, regulatory T cells, astrocytes and keratinocytes, where it shapes immune tolerance and central nervous system homeostasis. By controlling local TGF-beta availability, alpha V beta 8 restrains anti-tumor immunity, and blocking antibodies are in clinical trials to combine with checkpoint inhibitors in immuno-oncology.
