Description

[Pt(DACH)(OH)2(ox)] is a Pt(IV) derivative of Oxaliplatin (HY-17371) with two axial hydroxido ligands, used as a precursor for synthesizing Pt(IV)-HDAC inhibitor prodrugs. [Pt(DACH)(OH)2(ox)] exerts cytotoxic activity against human cancer cells and accumulates in cancer cells for nuclear internalization and nuclear DNA platination. [Pt(DACH)(OH)2(ox)] can be used for the research of lung carcinoma, breast carcinoma, pancreatic carcinoma, colon carcinoma[1].

In Vitro

[Pt(DACH)(OH)2(ox)] (Dihydroxy Oxaliplatin-Pt(IV)) (72 h) exhibits cytotoxic activity against MCF-7, A549, HCT-15, PC3, A498, BxPC3, and A375 cancer cell lines with an average IC50 of 13.2 μM[1].
[Pt(DACH)(OH)2(ox)] (24 h) does not inhibit HDAC activity in MCF-7 cells or isolated MCF-7 nuclei[1].
[Pt(DACH)(OH)2(ox)] (19.88 μM; 12-24 h) induces minimal nuclear DNA fragmentation in MCF-7 cells[1].

Result: Induced lower levels of nucleosome formation than bis-valproate and bis-phenylbutyrate cisplatin Pt(IV) derivatives (I and VI) and cisplatin.
Showed nucleosome formation levels only slightly above untreated control cells after 24 hours.
