Background

FGF-21, a stress-inducible hormone, regulates energy balance and glucose and lipid homeostasis through a heterodimeric receptor complex comprising FGFR1 and β-klotho. When binding to the FGFR1-β-klotho complex, FGF-21 triggers autophosphorylationof FGFR1 and activation of the downstream MAPK signalling cascade, and leads to phosphorylation-dependent activation of ERK1/2. FGF-21 can also signal through FGFR2 and FGFR3[1].
In additon, FGF-21 stimulates the oxidation of fatty acids, the production of ketone bodies, and the inhibition of lipogenesis. FGF-21 decreases blood glucose and plasma insulin in ob/ob obese mice, and can be used for research of obesity, NASH, NAFLD, and type 2 diabetes mellitus[1][2].
