Relatlimab

Human IgG4 kappa

Flow cytometric analysis of 1X10⁶ Jurkat cells with Relatlimab (HY-P99156, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

Loaded Relatlimab on AHC2 biosensor, can bind LAG-3 Protein, Human (HEK293, His, HY-P70722) with an affinity constant of 8.814E-10 M as determined in BLI assay.

In the presence of HLA-DR1 HA_306-318 aAPC, Relatlimab (anti-LAG-3) (20 μg/mL; 75 μL; 30 min; 37 ℃) reversed the LAG-3 overexpression-induced suppression of GFP reporter signal in human LAG-3-transduced Jurkat NFAT-GFP reporter T cells.
