Description

Rovunaptabin (ARC 183) is an aptamer, a single-stranded DNA molecule composed of 15 deoxynucleotides. It acts as a broad-spectrum neutralizer of pathogenic G-protein-coupled receptor autoantibodies. Rovunaptabin can be used in research related to cardiomyopathy and pulmonary hypertension[1][2].

In Vitro

Rovunaptabin (100 nM; 1 h) shows species-specific neutralization in spontaneously beating neonatal rat cardiomyocytes, fully neutralizing rat and rabbit G-protein-coupled receptor autoantibodies, partially neutralizing goat autoantibodies, and failing to neutralize chicken autoantibodies[1].
Rovunaptabin (100 nM) binds to the Fab region of human ETA-receptor and beta1-adrenoceptor autoantibodies, preventing restoration of agonistic activity via cross-linking in spontaneously beating neonatal rat cardiomyocytes[1].
Rovunaptabin (1 μM) forms a detectable complex with 1.5 μM human beta1-adrenoceptor autoantibodies in an electrophoretic mobility shift assay[1].

In Vivo

Rovunaptabin efficiently neutralizes beta1-adrenoceptor autoantibodies in vivo in spontaneously hypertensive rats and clears both beta1-adrenoceptor and muscarinic M2-receptor autoantibodies from rat serum[1].
Treatment with Rovunaptabin (BC 007) (1-4 mg/kg; i.v. infusion over 20 min; single dose; second single dose (9 months after initial)) safely neutralizes β1-adrenergic receptor autoantibodies in β1-AAB-positive Doberman Pinschers with dilated cardiomyopathy, reduces or stabilizes left ventricular volumes during β1-AAB-free periods, and significantly prolongs median cardiac survival to 572 days[2].
