Description

SP11 is a mitochondrial fission protein 1 (Fis1) inhibitor with an IC50 of 9.4 μM. SP11 binds only to activated Fis1 by engaging Cys41. SP11 preserves mitochondrial integrity and function during oxidative stress, inhibits hydrogen peroxide-induced mitochondrial ROS production, mitochondrial fragmentation, and Drp1 mitochondrial translocation. SP11 can be used for the research of parkinson’s disease[1].

In Vitro

SP11 potently and specifically inhibits to the Cys41 residue of activated Fis1Thr34Asp, with an IC50 of 9.4 μM[1].
SP11 (250 nM; 24 h) prevents hydrogen peroxide-induced mitochondrial ROS production, fragmentation and increased Drp1 translocation to the mitochondria in HK-2 cells[1].

Western Blot Analysis[1]

Cell Line: HK-2 human renal proximal tubule epithelial cells
Concentration: 250 nM (pre-incubation, then co-treated with 100 μM hydrogen peroxide)
Incubation Time: 30 min (pre-incubation); 24 h (co-treatment)
Result: Significantly reduced hydrogen peroxide-induced Drp1 recruitment to mitochondria.
Lowered Drp1/VDAC levels to near vehicle control levels.
