Surzebiclimab (BGB-A425) is a humanized IgG1-variant monoclonal antibody against T-cell immunoglobulin and mucin-domain containing-3 (TIM-3). Surzebiclimab binds to the extracellular domain of human Tim-3 with high affinity (KD=0.36 nM) and specificity. Surzebiclimab can be used in research of cancer.

Surzebiclimab (BGB-A425; 0.1-100 μg/mL) efficiently blocks the interactions between Tim-3 and PtdSer. Surzebiclimab enhances IFN-γ production of primary T cells and NK-mediated cytotoxicity against tumor cells[1].
Surzebiclimab augments T-cell response to allogeneic antigens either alone or in combination with an anti-PD-1 antibody BGB-A317. Surzebiclimab can also induce the internalization of Tim-3 receptor on cell surface[1].

Flow cytometric analysis of 1.5X106 MCF-7 cells with Surzebiclimab (HY-P99962, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. AF488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control.
